RCC1 is a chromatin-associated guanine nucleotide exchange factor (GEF) for the small GTPase Ran. RCC1 and Ran play key roles in nucleo-cytosolic protein transport and nuclear envelope assembly during mitosis. RCC1 is a WD40 domain containing protein and also a homolog of plant UVR8. Our SAXS data highlight the biological monomer of RCC1.
Data were prepared at 5 mg/mL and then diluted by 2/3 dilution series for a total of 4 samples. X-ray exposure data from 0.5 to 5 seconds for the ~1.5 mg/mL sample were used for further analyses.

The Guinier plot is used to estimate the radius of gyration, Rg, which is taken from the slope of a line observed at low scattering angles (typically in the range where q* Rg < 1.3). This should be in reasonable agreement with the real space Rg.


Kratky Plot

The Kratky plot can be used to visually assess the degree of "unfoldedness" of a protein or RNA sample. The plot of a well-behaved folded protein approaches the baseline at high q values creating a parabolic shape.

Description

RCC1 is a chromatin-associated guanine nucleotide exchange factor (GEF) for the small GTPase Ran.
