Scattering data were collected from a 1.5 mm glass capillary at 10°C under continuous flow (0.2 ml/min) with 2 second frames. hIL-11 (10 mg/ml) was eluted directly into the capillary from an in-line Superdex 200 5/150 size-exclusion column.

The Guinier plot is used to estimate the radius of gyration, Rg, which is taken from the slope of a line observed at low scattering angles (typically in the range where q* Rg < 1.3). This should be in reasonable agreement with the real space Rg.


Kratky Plot

SAXS data were acquired to complement the crystal structure and analytical ultracentrifuge analysis of hIL-11. Solution studies closely match our crystallographic model and suggest a stable monomeric structure for hIL-11.

The Kratky plot can be used to visually assess the degree of "unfoldedness" of a protein or RNA sample. The plot of a well-behaved folded protein approaches the baseline at high q values creating a parabolic shape.
